The experiment was conducted to determine the glucose concentration of solution using spectrophotometer. A spectrophotometer measures the absorption of light of specific wavelengths by a solution and allow us to determine concentration of glucose in the solution. In this experiment, a glucose solution was diluted into a few samples containing different glucose concentration. Different concentration of glucose in the solution show a distinct result of absorbance of light in the spectrophotometer. Therefore, we can assume that analysis using absorbance at specific wavelength of lights can help to determine the glucose concentration in solution. Once we plot the absorbance versus glucose concentration, we can easily determine the concentration of glucose containing in the solution. From the graph, the linear relationship between absorbance and glucose concentration can be identified. Some precaution step should be carry out during the experiment. The solution that we measure contains the substance that we want to measure, along with solvents. These solvents may absorb light as well. To prevent this, a blank sample has been used to calibrate the spectrophotometer to attain the zero concentration. This is important to allow us to obtain an accurate absorbance value of light of a sample.
